目的 探讨MMPs抑制剂联合环磷酰胺(CTX)干预对结肠癌细胞生物学特性及几丁质酶样蛋白-40(YKL-40)和人类表皮生长因子受体-2(HER-2)表达的影响。方法 人结肠癌细胞株Lo Vo根据实验设置分组为4组:空白对照组(进行溶剂二甲亚砜干预)、CTX干预组(仅进行2μmol/L CTX干预)、GM6001干预组(仅进行2.5μmol/L GM6001干预)和联合干预组(2μmol/L CTX和2.5μmol/L GM6001共同干预)。干预24 h后,采用Realtime PCR检测细胞YKL-40和HER-2 mRNA表达;干预48 h后,蛋白质印迹法检测细胞凋亡蛋白表达;干预72 h后,酶联免疫吸附试验法试剂盒检测细胞上清中YKL-40和HER-2水平;分别于干预24 h、48 h和72 h后,MTT细胞增殖检测试剂盒检测细胞增殖能力。结果 与空白对照组相比,CTX干预组、GM6001干预组和联合干预组细胞YKL-40和HER-2 mRNA和蛋白相对表达量均显著降低,联合干预组也显著低于GM6001ventriculostomy-associated infection干预组和CTX干预组,差异均有统计学意义(P <0.diABZI STING agonist体内实验剂量05),但GM6001干预组和CTX干预组相比差异无统计学意义(P> 0.05)。与空白对照组相比,CTX干预组、GM6001干预组和联合干预组细胞抗凋亡蛋白Bcl-2相对表达量显著降低,促凋亡蛋白Bax和cleaved-Caspase-3相对表达量均显著增加,差异均有统计学意义(P <0.05);联合干预组Bcl-2表达也显著低于GM6001干预组和CTX干预组,Bax和cleaved-Caspase-3表达也显著高于GM6001干预组和CTX干预组,差异均有统计学意义(P <0.05),但GM6001干预组和CTX干预组凋亡蛋白表达相比差异无统计学意义(P> 0.05)。干预24~48 h后,空白对照组、CTX干预组和GM6001干预组细胞增殖能力均随时间增加而显著上升,差异均有统计学意义(P <0.05),而联合干预组不同时间差异无统计学意义(P> 0.05);干预不同时间点,与空白对照组相比,CTX干预组、GM6001干预组和联合干预组细胞增殖能力均显著降低,联合干预组也显著低于GM6001干预组和CTX干预组,差异均有统计学意义(P <0.05),但GM6001干预组和CTX干预组相比差异无统计学意义(P> 0.05)。结论 MMPs抑制剂联合环磷酰胺干预结直肠癌细胞可通过促进细胞中促凋亡蛋白表达、抑制抑凋亡蛋白蛋白,从而促进结直肠癌细胞凋亡,并可抑制细胞增殖,因此可能进一步抑制结直肠癌发展和恶化,其作用机制可能与抑制细胞中YKL-40和HER-2的表达此网站相关。
